RAPD Analysis of an Easter Lily Chlorophyll Mutant

A white edged leaf strip chimera mutant was obtained by in vitro mutation breeding. The flower bud was excised from the mutant and underwent tissue culture regeneration in an effort to isolate pure mutations. Three types of leaf color (white, yellow, and light green) shoots were regenerated from the flower bud; however, only light green color shoots rooted in rooting medium and were transferred to soil for greenhouse testing. Chlorophyll content and random amplified polymorphic DNA (RAPD) were evaluated between the regenerated green mutant plant and the control plant. The mutant plant had significantly lower chlorophyll content than the control, and DNA marker analysis showed clear DNA level differences, meaning a chlorophyll lacking mutant was created in Easter lily.
