Lily hybrids originated from oriental-trumpet hybridization, also called orienpets or OT hybrids. These interspecific hybrids produce lily bulbs that easily endure late season frosts without bud kill. These reliable plants are known for their sweet fragrance, a shape unique to Oriental lilies, and increased drought-resistance. Traditional hybridizers are working to increase the color range of orienpets, and farmers are testing efficient propagation methods to make limited varieties available for distribution. Plant tissue culture based in vitro technology can be a tool for orienpet lily biotech breeding and novel variety propagation. We established a callus culture plant regeneration system for orientpet lily. Callus was induced from bulb scale of orienpet lily ’Silk Road’ on MS medium supplemented with various levels of NAA. Embryogenic callus was obtained through continuous selection of callus, milk-yellow in color, from MS medium with NAA 2mg/L for three months. Plantlets regenerated from embryogenic callus on NAA level reduced MS medium. Charcoal added (1mg/L) MS medium was used for regenerated plantlet strength and rooting. Flowering of callus regenerated plants showed no somaclonal variation in greenhouse.
